The aim of the investigation: to evaluate the influence of smoking parents on the endothelium of blood vessels of descendants, using experimental modeling of smoking parents.
Material and methods. Formed «couples»: in a special chamber with tobacco smoke for 15 minutes every day: 1) «smoked» only females(mother before mating 1month and during gestation; 2) «smoke» is only males-fathers 1month before mating; 3) «smoked» males and females, ie, both parents; 4) a control group of rats — intact. In each group 15 newborn animals were studied: gr.M, gr.F, gr.MF, gr.C. In the blood serum endothelin-1, vascular endothelial growth factor (VEGF) by smmunoassay methods using ELISA kits of DRG firm reagents (Germany), and S-nitrosothiols (stable NO metabolites) by spectrofluorimetric method were studied. In sections of brain tissue immunohistochemical reaction to 8 coagulation factor was supplied. We used antibodies firms Cell Marque (USA) with its visualization by peroxidase. Within photograph microscope slide (x400) the length of the capillary walls, as well as absolute and relative length of the deendotelizated capillaries walls were determined.
Results. Blood concentrations of endothelin(1 in the main groups are increased, and S-nitrosothiols — are reduced, the ratio E/S-NO in all main groups are increased significantly: K<F<M<MF. Concentrations of VEGF in the blood serum of the main groups are reduced (except females of gr.F). Immunohistochemistry were revealed that the length of deendotelizated capillary walls in the main groups is increased (more than 50%, whereas s gr.C — about 20%).
Conclusion. In the experiment, we found increased endothelial vasoconstrictor potential, decreased endothelial resistance to damaging factors and the reduction of the regenerative capacity of endothelium.
