The objective: to study the frequency of genotypes and allelic variants of the IL-1a (-889 C>T), IL-1b (3953 C>T) genes, depending on the reproductive status and to evaluate association with recurrent miscarriage, which set in cycles of in vitro fertilization (IVF), from residents of Odessa region of Ukraine.
Patients and methods. We observed 240 patients with recurrent miscarriage, came in IVF cycles, and 100 apparently healthy fertile women of the control group with a history of at least one term delivery and lack of spontaneous abortion episodes. For typing single nucleotides polymorphisms of immune responses genes IL-1a -889 C>T and IL-1b 3953 C>T polymerase chain reaction is used to melting the reaction products in the presence of «adjacent» oligonucleotides.
Results. ТТ-889 genotype of gene IL-1a (OR 6.86; 95% CI: 2.07–22.69) and ТТ3953 genotype of gene IL -1b (OR 9.22; 95% CI: 2.18–38.99) were associated with the possible increase in the risk of recurrent miscarriage after IVF.
Conclusions. Typing IL1a -889 C>T and IL1b 3953 C>T gene SNPs immune response can be used as a method of early diagnosis and prediction pregravidal reproductive losses in infertile women after IVF. The ТТ-889 genotype of IL-1a gene and ТТ3953 genotype of IL-1b gene associated with the possible increase in the risk of recurrent miscarriage after IVF.
Key words: fertilization, recurrent miscarriage, genotype, gene polymorphism, IL-1a, IL-1b, Odessa region.
